Gut Microbe Breakthrough: KIST Researchers Discover Natural Compound to Combat Deadly Cancer Cachexia

SEOUL — Up to 80 percent of cancer patients face a debilitating complication known as cancer cachexia, a severe metabolic syndrome characterized by rapid weight loss and muscle wasting. Associated with 20 percent of all cancer-related deaths, the condition currently lacks fundamental pharmacological treatments. However, a groundbreaking discovery by researchers at the Korea Institute of Science and Technology (KIST) offers a promising new therapeutic strategy using a natural compound that targets the gut microbiome.
Announced on August 16, a joint research team led by Dr. Myung-seok Kim and Dr. Choong-gu Lee at KIST’s Natural Product Efficacy Optimization Research Center screened a library of over 4,000 natural products. They discovered that micheliolide (MCL), a plant-derived compound known for suppressing inflammation and regulating protein modification, effectively ameliorates cachexia.
In animal models of colorectal and pancreatic cancer, MCL administration increased muscle mass by 34 percent and muscle strength by 31 percent. Crucially, the compound achieved this without directly affecting the tumor size, successfully uncoupling muscle preservation from tumor progression. Furthermore, MCL restored impaired T-cell immune functions and suppressed the secretion of inflammatory cytokines that induce muscle breakdown.
The research team found that this anti-cachectic efficacy is intrinsically linked to the gut-immune-muscle axis. In mice treated with antibiotics to deplete their gut microbes, MCL’s muscle-rescuing effects vanished. Subsequent analysis revealed that MCL selectively enriches Phocaeicola vulgatus, a beneficial gut bacterium.
Taking the research a step further, scientists investigated the metabolites produced by this specific bacterial strain and successfully identified 11 new cyclopeptides directly involved in promoting muscle synthesis and suppressing systemic inflammation.
Published in the prestigious international journal Microbiome in May 2026, these findings present a highly translational, host-directed intervention.
"This research is significant as it demonstrates the possibility of ameliorating cancer cachexia through the interaction between natural compounds and the gut microbiome," said Dr. Kim. "Moving forward, we will strive to develop innovative new drugs—potentially as accessible oral therapeutics—through further clinical research and technology transfer to increase the survival rate and alleviate the treatment burden for cancer patients."
